I have changed the email tracking token prefix in our test tenant, and would like to test it before making the same change in the live tenant. As the test tenant is not connected to the email router I am struggling with how to test the change. Does anyone have any suggestions please?

I guess you could configure email going out using outlook client and then test this functionality ..
so for one user (may be your self or test user) install CRM client, go to CRM user account and then set Email access configuration out going and incoming as outlook..now this user will able to send email from CRM..
send one email from this user and check email at receiving end for this configuration check..
