Are you referring to Windows Live Mesh or Live Mesh beta - www.mesh.com? Since you mention the Notifier, it sounds like you are still using Live Mesh beta.
Basically, you need to go to the device you are looking to connect *to* and enable remote connections. Using Windows Live Mesh, open it via the icon in the task bar, and click on Remote. You will see the option to allow or block connections. Blocking is the default. If you have enabled remote connections, and continue to get the error, your firewall is probably blocking the connection.
If you are referring to Live Mesh beta, note that it will cease to function in a few weeks - end of March - so you should uninstall it and install Windows Live Mesh from http://download.live.com
