Quickest is that 70-528 is for .NET 2.0 and 70-562 is for .NET 3.5
The best way for you to make this decision is which framework do you work in daily and go for that exam. You will also need to do 70-536 exam as well to get the MCTS

70-528 for .net 2.0 ,and 70-562 for .net 3.5 .your question is equal to what technology is more suitable for you ,and or will give you a promising future.
3.5 is that it's only fully supported in VS2008 so you'll need to have a copy of that (or the VS2008 express editions) to make best use of it.besides .3.5 is the latest technology and
3.0 was that it provided windows presentation foundation, communication foundation, workflow foundation and cardspace, but that sits on top of the 2.0 framework.
.net 3.5 vs .net 2.0 exams just like server 2008 exams vs server 2003 exams or windows 7 exams vs vista exams .
The old technology is the foundation of the latest technolgy.
If you pass .net 2.0 exams or get .net 2.0 cert ,if it is necessary ,you can consider upgrade .net 2.0 cert to .net 3.5 certs ,which will be easier after your passing .net 2.0.
Besiders ,In my opinion,if you want to go to web-development in c# using asp.net ,you can consider .net 2.0 exams ,which more suitable for u.
Of course ,Niall's advise is also useful for u.you should consider about your working enviorment .
ps :your questions, to some extend, seems like to ask you which chewing gum tastes better: mint or lemon.however ,i have done my best to answer your questions ,hope it useful for u.

As far as I know,70 -528 cover many topics covered in 562,however ,70 -562 Pro ASP.net 3.5 in C#,most of the latter's contents focusing more on Linq ,ajax and Silverlight.
More information, maybe Net 2.0 70-528 better than .net 3.5 70-562 will help you .This is what I can do for u.
Hope it useful for u.

Ok from having completed both exams here is what I can tell you
There are some topics that are common to both exams such as web site configuration, security using the web.config, master pages and user controls
In 2.0 there is no AJAX questions as the libraries were not shipped out of the box in ASP.NET 2.0. Additionally there are no questions on WCF or Silverlight. It covers a lot of the differences between ASP.NET 1.1 and 2.0
In the 3.5 exam you can expect questions on the out of the box AJAX functionality, some WCF questions (calling from within ASP.NET and ASP.NET AJAX), LINQ and I dont think there is Silverlight again as this was not out of the box functionality and has gone through 3 or 4 revisions in a very short space of time.
Will you be lacking in some skills on choosing 3.5 or 2.0.. no not really, as the CLR for both versions is the same and the main concepts havent changed between 2.0 and 3.5.

Here is what i learned from ur posts.......
Exam 70-562 is for .NET 3.5 it is latest web development
(http://www.microsoft.com/learning/en/us/exam.aspx?ID=70-562&locale=en-us#tab2)
Exam 70-528 is for .NET 2.0 is old web development
http://www.microsoft.com/learning/en/us/exam.aspx?ID=70-528&locale=en-us#tab2
So, are the skills measured for both the certification same or will i be lacking some skills if i select Exam 70-562 over Exam70-528
