Hi, I had the same problem yesterday and found, thanks to posts here, that it was due to Avast! version 6 causing Vista to go into tamper mode.
To rectify it, assuming you use Avast! you'll need to remove it completely from your system. If you don't have access to your control panel then reboot into Safe Mode (F8) and then you should have access back. Remove Avast! and then download the uninstaller to remove all traces http://www.avast.com/uninstall-utility
Reboot twice and you should be fine. You may need to manually re-validate Windows www.microsoft.com/genuine/validate
