If the D: drive is a second internal drive, it should be available to OneCare's backup settings and it should be scanned for malware and Defrag will also make a pass at it during Tune-up. There is no configuration for Tune-up other than the schedule.
If D: is an external drive, it can't be backed up, nor will a Defrag execute on it. You'd have to do the latter manually via Start/All programs/Accessories/System Tools/Disk Defragmenter
