Hi, Kevin.
There have been a few reports of this issue in the past few days and I'm wondering if there is any relationship to the problems that began on the 11th of July for some users.
If a reinstall didn't resolve it, then I suggest opening a Support case for help:

However, I would personally use the opportunity to seek a different backup solution right now since OneCare is at End of Life.
See this post for End of Life information for Windows Live OneCare - http://social.microsoft.com/Forums/en-US/onecaregeneral/thread/81996436-edad-41ba-bdfc-02fc094e2ca9
You can uninstall OneCare http://social.microsoft.com/Forums/en-US/onecaregeneral/thread/732e6371-2e71-479a-8cd6-b189941ed4fb, use the cleanup tool: http://social.microsoft.com/forums/en-US/onecareinstallandactivate/thread/399071e2-49dc-4767-932d-d261e3965943/, and then download and install the free Microsoft Security essentials for malware protection - http://www.microsoft.com/security_essentials/resources.aspx - or you can wait until your subscription expires, which will be by the end of 2010 at the latest.
If you used OneCare for backups, you'll still be able to restore from old backups if you use the OneCare Restore tool - http://social.microsoft.com/Forums/en-US/onecarebackupandrestore/thread/e5d686c8-762b-4b22-bc08-bfb93f911e57, but you'll need to seek an alternative backup solution.

If you performed a System Restore wait a day or two then try to configure backup. If the issue persists you can contact support for help. How to reach support - http://social.microsoft.com/Forums/en-US/onecareinstallandactivate/thread/30400b52-7f26-4ba0-bc18-17e305329d90

I have Windows Live OneCare installed, and everything seems to be working except Backup and Restore. My Windows Live OneCare icon is yellow and when I click it, the Windows Live OneCare opens with a button labeled "Configure backup". When I click the "Configure backup" button, I get a message "Backup Cannot Start. OneCare is initializing. Please try again later." The only option is a "Close" button. This message has been popping up for about a week now -- even after the computer has been turned on for a few hours.
How can I get OneCare initialized so I can configure my backup and start backing my stuff up?
Thanks. Bill

personally, I'd skip the backup and start looking for an alternative backup solution since OneCare is at end of life.

However, if you want to try to resolve this and configure backup, I recommend contacting support.

Steve,
Thanks for the response. Didn't realize OneCare was near end of life. How long will it be useful? Will there be a replacement? What is recommended?
Thanks. Bill

I've just merged multiple threads into a single discussion. I also deleted some replies to remove duplication.
I'm contacting Microsoft about this issue as it does appear that more than a few people are having the problem - it seems like an account/back end issue at this point.
