This is a complete beginner question, so I apologize if it is misplaced, but can CRM be deployed on 3 tiers (UI separate to web services)?

With Enterprise edition you have several server roles.

I recommend the "Implementation Guide"

Absolutely. In theory you could create a completely new GUI and have it chat to the web services.
